研究室簡介

紅龍果病毒之鑑定、檢測以及交互作用之研究

仙人掌科三角柱屬的紅龍果為國內發展迅速之高價熱帶水果,除1999年本系劉瑞芬教授實驗室首先發現的仙人掌X病毒(Cactus virus X, CVX)外,本實驗室亦從紅龍果分離出蟹爪蘭X病毒(Zygocactus virus X, ZyVX),以及未曾被鑑定之新potexvirus-紅龍果X病毒(Pitaya virus X, PiVX)。我們針對這三種potexviruses開發multiplex RT-PCR以及magnetic nanoparticle-capture RT-PCR (MNC RT-PCR),可確認樣品內病毒的種類。利用這兩種檢測方法我們曾進行兩次全省調查,發現國內紅龍果園多已遭受這三種potexviruses所危害,且多為複合感染。

我們已獲得CVX和PiVX感染性cDNA株,目前還在構築ZyVX的cDNA株。因此目前先以原生質體和植株分析CVX和PiVX之複製和移動能力,研究病毒間的交互作用,以及對寄主植物之影響。

另一方面,我們希望利用之前所研發的檢測方法,配合農業研究單位和農民,篩選無病毒材料;同時嘗試開發紅龍果去病毒技術,藉以建立無病毒紅龍果種苗生產體系,提昇我國紅龍果產業之國際競爭力。


花卉病毒之研究與檢測方法之研發

人工栽培之蘭花常有蕙蘭嵌紋病毒(Cymbidium mosaic virus, CymMV)和齒舌蘭輪斑病毒(Odontoglossum ringspot virus, ORSV)複合感染,且導致植株病徵加重的協力現象發生。本實驗室發現CymMV與ORSV同時感染圓葉菸草(Nicotiana benthamiana)時亦有協力現象,因此希望從複製、移動、基因靜默等不同面向,探討CymMV與ORSV協力作用之機制。

對危害蘭花的兩種重要病毒:蕙蘭嵌紋病毒(Cymbidium mosaic virus, CymMV)和齒舌蘭輪斑病毒(Odontoglossum ringspot virus, ORSV),研發多種專一且快速的檢測方法,包括ELISA、tissue blot immunoassay、multiplex RT-PCR、immunocapture RT-PCR與基因晶片等,除了供教學和學生實驗使用,亦可應用於我國文心蘭與蝴蝶蘭種苗病毒驗證作業。


重要植物病毒快速檢測和鑑定系統之建立

針對我國有害生物名單中之重要植物病毒建立鑑定相關資料庫與標準鑑定流程,提供植物檢疫使用。

針對potyvirus研發廣效性單株抗體與ELISA檢測法、屬專一性引子對與RT-PCR檢測法,以及核酸探針、基因晶片等快速檢測和鑑定系統。
