研究室簡介

大豆抗病遺傳與病害防治研究

大豆 (Glycine max (L.) Merr.) 是臺灣重要作物之一,其使用途徑可二分為飼料油用的穀粒大豆或俗稱黃豆(grain soybean),以及作為蔬菜食用的毛豆或俗稱枝豆 (vegetable soybean, or edamame)。其中毛豆的外銷價值於近年來穩定突破二十億臺幣,故又被稱作「臺灣綠金」。黃豆與毛豆雖有不同的農藝育種偏好,但抗病蟲害之特性是黃豆和毛豆共同的目標。其中種子與幼苗期的根腐病是兩者皆常遇到的病害,嚴重時可造成種子腐爛或幼苗萎凋。根腐病的防治仰賴農藥於種子披衣 (seed coating),但由於根腐病可由五種病原菌造成(Fusarium root rot, Macrophomina root rot, Phytophthora root rot, Pythium root rot, Rhizoctonia root rot),且已有不同病原菌之抗藥性形成,因此除了種子披衣之外,篩選抗病品系以及研究抗病基因為另一重要之病害防治方法。

我們實驗室研究台灣毛豆各式病害,方向包括分離重要之病原菌、篩選抗病性、以及利用 Genotyping-by-Sequencing (GBS)、QTL Mapping、Genome-wide Association Study (GWAS)等基因定位與統計方法,尋找與抗病關聯之分子標記與基因,期望提供台灣毛豆對於病害防治的學理與應用基礎。

大豆紅冠腐病之防治藥劑篩選研究

博士班吳秉祜同學應用全光譜平面掃描技術,建立了高通量的藥劑篩選平台,並納入 Z' factor 統計分析以提升高通量藥劑篩選的可信度。本研究亦與高雄改良場曾敏南課長、屏東科技大學植物醫學系林盈宏教授、以及嘉義大學植物醫學系郭章信特聘教授合作研究,收集了多年的大豆紅冠腐病菌株,並在篩選多種藥劑與評估族群抗藥性之後,發現賽普護汰寧於溫室接種下展現最佳的保護效力。未來將持續研究賽普護汰寧以種子披衣於田間防治大豆紅冠腐病之潛力。相關研究成果已發表在美國植物病理學會的Plant Disease期刊。

根圈細菌誘導大豆幾丁質酶參與抗病反應之研究

碩士班陳政諺同學針對三個演化相近但基因差異表現的大豆幾丁質酶基因,與中研院吳志航博士合作利用阿拉伯芥基因轉殖技術進行驗證。結果發現雖然過表達其中二個幾丁質酶基因可降低鐮孢菌造成之病徵,但兩者在受到伯克氏菌誘導之基因表現上已有分歧。另以六種菌屬的根圈細菌進行轉錄體分析,觀測到大豆基因體內的幾丁質酶對不同菌屬的誘導反應不同,且能夠被誘導表現的幾丁質酶啟動子區間,偵測到與根瘤發育相似的保守性序列模體,顯示不同根圈細菌在誘導大豆幾丁質酶基因表現上已有演化專一性及多元化之現象。相關研究成果已發表在Frontiers in Plant Science期刊。

集群分離RNA測序分析於大豆白粉病之抗病定位研究

碩士班黃承濬同學應用集群分離RNA測序分析(Bulk Segregant RNA-Seq, BSR-Seq),針對由桃園改良場林楨祥研究員所建立的白粉病抗病品種「高雄11號」與感病品種「花蓮一號」雜交族群進行QTL定位。根據∆(SNP-index)與G'兩種方式所計算之QTL交集,搭配以應用等位基因專一性引子對所預測之族群抗病性,結果顯示在大豆第16號染色體上的Rmd基因座是「高雄11號」的主要抗病性來源。本研究首次在大豆物種上證實BSR-Seq方法的適用性,並解開「高雄11號」的抗病遺傳組成。相關研究成果已發表在BMC Plant Biology期刊。

 

 

 

 

 

 

 


菌核生物學

核盤菌(Sclerotinia sclerotiorum)、炭腐病菌(Macrophomina phaseolina)、立枯絲核菌(Rhizoctonia solani)、白絹菌 (Athelia rolfsii) 等植物病原真菌可以形成菌核或微菌核等特化構造,在田間殘存並成為未來的感染源。因此研究菌核的形成機制,達到降低菌核於田間殘存的目標,是我們實驗室的研究興趣,並期望能針對形成菌核的病原真菌,開發整合性病害防治策略。

炭腐病菌之微菌核形成機制研究

碩士班劉軒豪同學以炭腐病菌的微菌核為研究材料,建立了四個菌核形成階段的組織萃取方式,並利用高通量定序及轉錄體方式,發現炭腐病菌的微菌核形成與活性氧(Reactive oxygen species, ROS)相關基因表現有顯著相關。後續在抗氧化劑與氧化刺激物的外源添加培養實驗中,深入發現O2-扮演了刺激菌絲分化成微菌核的重要關鍵角色。相關研究成果已發表在美國微生物學會的Microbiology Spectrum期刊。

立枯絲核菌AG-7之基因體與菌核遺傳研究

碩士班林鈺晟同學以立枯絲核菌AG-7為研究材料,以Oxford NanoPore及Illumina Sequencing完成了全球第一個AG-7菌絲融合群的基因體;另應用測序基因分型(GBS)鑑定154株分離自台灣的立枯絲核菌族群,並以全基因體關聯分析(GWAS)定位與菌核數量及菌核大小連鎖的SNPs。分析結果顯示立枯絲核菌的菌核形成能力應為可遺傳的性狀,其中與菌核數量連鎖的基因體區間有顯著富集的活性氧(ROS)相關基因功能。相關研究成果已發表在英國微生物學會的Microbial Genomics期刊。
